Proliferating cell nuclear antigen (PCNA) plays essential roles in DNA metabolic processes including replication, repair, recombination and cell cycle progression. PCNA forms a trimeric ring that encircles duplex DNA and associates with, and influences the activity of, many proteins participating in DNA metabolic processes and cell cycle progression. We study the two PCNA homologues from the thermophilic archaeon Thermococcus kodakarensis. We have solved the three dimensional structure of the proteins and characterized their biochemical properties. A small protein that binds to PCNA and regulates its activities has also been identified. These studies will be presented.
