Imaging of the role of dopamine in behavioural disorders

We often need to adapt behaviour to changing circumstances - to plan novel actions or inhibit pre-potent ones. These decision making process depends on the accurate representations of reward or losses and the inhibition of impulses. Abnormalities in this process can lead to impulse control disorders. In Parkinson´s disease (PD), the decision of which action to take, and the inhibition of harmful behaviour, can be impaired and further worsened by dopaminergic therapy. The combination of PET and fMRI is revealing the mechanisms of poor impulse control in PD. Here, we show the results of neuroimaging studies conducted in PD patients with and without ICDs providing evidence of significant abnormalities in those prefrontal-striatal areas associated with decision making and reward. Proposed mechanisms include abnormal functioning of mesolimbic structures resulting in dysregulation of dopamine.
