Smallpox has been one of the most devastating diseases of humanity. Working on the safe model system vaccinia virus we study the heart of DNA replication formed by the polymerase holoenzyme consisting of the polymerase E9, the uracil-DNA glycosylase D4 and a bridging protein A20 on one hand and the hexameric helicase-primase D5 on the other hand. Combining biophysical methods (such as crystallography, electron microscopy, SAXS, MALLS) in a multi-scale
approach we progress towards the structure, and using for example surface plasmon resonance and fluorescence anisotropy, towards the function of the machinery.
