The “contact phase”, or kinin–kallikrein system, consists of blood proteins that play a role in inflammation, innate immunity, blood pressure control, coagulation and pain. Its important mediators, bradykinin and desArg9-bradykinin peptides, develop vasopermeability and vasodilatation with high affinity receptors. The system consists of proenzymes that can be activated upon triggers, with kininogens as substrates and small peptides as effectors ; this process is under a tight control by the serpin C1 Inhibitor. The major fundamental questions target the activation process, its control and ligand-receptor interaction. Our objective is to develop diagnostic tools appropriate to diseases that result from disequilibrium of the kinin–kallikrein system. Inflammatory diseases, e.g. angioedema or anaphylactoid reactions, are associated with an accumulation of kinin ligands on the B2 or B1 receptors. These conditions are dependent on an excess of kinin forming system or a defective kinin catabolism. Besides, iatrogenic conditions emerge with anti-hypertensive drugs, anti-diabetics, 5-alpha reductase inhibitors or mTOR inhibitors. The experiments are set up to decipher the enzymatic activities, the substrate cleavage with peptide production and subsequent receptor activation. Our research is translated to a Spin-off Company aimed, on one hand, to put forward our knowledge and expertise of the kinin forming system to help pharma companies in their decision of new medicines and, in the other hand, to solve the adverse effects of medicines such as anti-hypertensive drugs, haemodialysis, anti-diabetics, bloodderived
products.
