The role of the NF2/ merlin tumor suppressor gene in signal transduction, proliferation and cancer

The NF2 gene inactivation is responsible for a rare tumor predisposition syndrom called Neurofibromatosis type II. The patients develop multiple intracranial tumors arising from Schwann cells (schwannomas) and meninge(meningiomas). Conventionnal chemotherapy are ineffective and targeted treatment are virtually inexistant due to the lack of clearly identifiedtherapeutic targets. Our research focus is to get a better understanding into the mechanisms of the tumor suppressor function of merlin, the NF2 gene product as a mean to identify potential therapeutic targets . Our approaches combine Schwann cell biology and proteomic studies of human schwannomas. Our recent work links merlin to receptor tyrosine kinase turnover and to the regulation of the hippo signaling pathway in schwannomas suggesting that they both might represent valuable target for thearpeutic intervention.
