Protein structures : from spectroscopy to crystallography

X-ray crystallography is the technique of choice for obtaining high-resolution structural data on proteins today. For certain proteins, in particular those which absorb light in the visible range, optical spectroscopy can also provide high-resolution data on the structure of the visible-light absorbing chromophore and its protein environment. In this talk, I will use a few examples to demonstrate how optical spectroscopy can help to solve specific questions in structural biology, by itself and in combination with crystallographic data. Finally, I will discuss some aspects and challenges of future advancements regarding automation, sample handling, and data processing which are going to open new possibilities in protein crystallography.
