The dual role of the TNF ligand APRIL in tumor formation and inflammatory diseases

Altered expression or function of a Tumor Necrosis Factor (TNF) family member is frequently associated with disease development. Our group originally described that the the TNF ligand APRIL (A Proliferation-Inducing Ligand) promotes B cell chronic lymphoid leukemia (CLL) and, more recently, also colorectal cancer (CRC). Unexpectedly, we found that APRIL has also immunoregulatory capacities by stimulating regulatory B1 cells and, thus, dampening inflammation in various mouse models including collagen-induced arthritis. Following our finding that APRIL drives immunoregulatory B cells in mice, we study – in collaboration with the CHU Montpellier - whether APRIL could also promote human regulatory B (Breg) cells.
