Huntington’s disease (HD) is a widely studied disease that provides a model to understand brain response to proteotoxicity in neurodegenerative disease. An important question is to understand the rules that may govern the molecular dynamics of the HD process over time and how this may impact on neuronal function and synaptic homeostasis. Predictive modelling studies provide a new paradigm in which the alteration of cellular identity and capacity for stress resilience may cast the frame for the disease to develop in the adult brain.
