Viruses cause numerous diseases in plants, animals and humans. Characterizing the structure of the viral machineries involved in virus replication is clearly a major asset for understanding the mechanisms of viral infection and pathogenesis and for fighting against these pathogens. Recent studies revealed that disorder and
flexibility are widely spread features within the viral components, requiring the development of new approaches for their structural characterization. I will present some of our recent results concerning the structural and mechanistic characterization of the replication machinery of rhabdoviruses (rabies virus) and paramyxoviruses (measles virus, Nipah virus) and the structural flexibility of the
IRES RNA of the hepatitis C virus.
