Decision Making in the Cell Migration Process : Sensitivity to RhoGTPase Gradients

In order to navigate correctly, a cell needs to amplify some signals, to filter out other ones, and to take a decision based on a combination of them. The signaling network associated to RhoGTPases plays a key role in this process, as they play the dual role of integrating diverse environmental signals and actuating the cytoskeleton. Yet, the precise orchestration of RhoGTPase pathways in space and time is still poorly understood.
To get a quantitative understanding of the intracellular processes in charge of such tasks, we developed tools allowing the control of the cell microenvironment and allowing the perturbation of intracellular biochemical activities. Our work consists in dissecting the spatiotemporal regulation of the RhoGTPase signaling circuitry in order to understand how topology and coordination of these pathways provide sensitivity and robustness to the polarization process.
We use a controlled microenvironment modifiable chemically in order to initially deprive cells of any directional cue but let them migrate freely upon specific treatment. Using optogenetics and gradient-patterned light stimulation, we have developed a technique that allows to impose controlled gradients of RhoGEF concentrations across single cells. We measured various parameters of cell migration and observed different responses to cdc42 and Rac1 activity gradients. Importantly, we could show that a crosstalk between these pathways is necessary for the efficiency of cell navigation. We are currently investigating the topology of this crosstalk in order to understand how it affects the control of cell navigation during migration.
