Crystallography provides deep insight into the atomic composition of macromolecular compounds. It is not restricted to single molecules. Our understanding of of large complexes like the 14 subunit RNA-polymerase or the 80S ribosome was very much broadened when their crystal structures became available. Crystal structures are represented as colorful, exciting pictures. Biologists need to learn what type of information they can get from crystallographic data and, at least as important, what are the limits of crystallography. Great, very advanced techniques, based on crystallography and diffraction, are accessible for biologists nowadays. Data collection and analysis is often carried out by specialists, and only the results are presented to the biologist. With lack of understanding, the colorful pictures become prone to overinterpretation. This lecture is going to provide examples of what structural biologists can learn from a crystal structure and it aims to explain some of the limitations.The lecture will provide an overview of the individual steps from crystal growth via data collection to structure presentation.
